چکیده

Abstract The androgen receptor (AR) is the key oncogenic driver of prostate cancer, and despite implementation novel AR targeting therapies, outcomes for metastatic disease remain dismal. There an urgent need to better understand androgen-regulated cellular processes more effectively target dependence cancer cells through new therapeutic vulnerabilities. Transcriptomic studies have consistently identified lipid metabolism as a hallmark enhanced signaling in yet relationship between lipidome remains undefined. Using mass spectrometry–based lipidomics, this study reveals increased fatty acyl chain length phospholipids from patient-derived explants one most striking changes metabolism. Potent direct AR-mediated induction ELOVL acid elongase 5 (ELOVL5), enzyme that catalyzes elongation, was demonstrated cells, xenografts, clinical tumors. Assessment mRNA protein large-scale data sets revealed ELOVL5 predominant expressed upregulated compared with nonmalignant prostate. depletion markedly altered mitochondrial morphology function, leading excess generation reactive oxygen species resulting suppression cell proliferation, 3D growth, vivo tumor growth metastasis. Supplementation monounsaturated cis-vaccenic acid, product reversed oxidative stress associated proliferation migration effects knockdown. Collectively, these results identify elongation protumorigenic metabolic pathway androgen-regulated, critical metastasis, targetable via ELOVL5. Significance: This identifies phospholipid action

Fatty acid elongase-5 (Elovl5) regulates hepatic triglyceride catabolism in obese C57BL/6J mice.

Nonalcoholic fatty liver disease is a major public health concern in the obese and type 2 diabetic populations. The high-fat lard diet induces obesity and fatty liver in C57BL/6J mice and suppresses expression of the PPAR-target gene, FA elongase 5 (Elovl5). Elovl5 plays a key role in MUFA and PUFA synthesis. Increasing hepatic Elovl5 activity in obese mice lowered hepatic TGs and endoplasmic r...

the relationship between polyunsaturated fatty acid intake and prostate cancer

prostate cancer is the most common cancer in america and canada, and is the sixth most common cancer worldwide. dietary factors play an important role in prostate cancer development; one of them is dietary fat, particularly pufa intake. thus, this study aimed to review the current articles on the correlation between pufa and prostate cancer. we searched the pubmed database, and selected article...

Dual Fatty Acid Elongase Complex Interactions in Arabidopsis

Very long chain fatty acids (VLCFAs) are involved in plant development and particularly in several cellular processes such as membrane trafficking, cell division and cell differentiation. However, the precise role of VLCFAs in these different cellular processes is still poorly understood in plants. In order to identify new factors associated with the biosynthesis or function of VLCFAs, a yeast ...

Interleukin-6: A multifunctional targetable cytokine in human prostate cancer

Several cytokines are involved in regulation of cellular events in prostate cancer. Interleukin-6 (IL-6) was frequently investigated in prostate cancer models because of its increased expression in cancer tissue at early stages of the disease. In patients with metastatic prostate cancer, it is well-known that IL-6 levels increase in serum. High levels of IL-6 were measured in the supernatants o...
